POS0975 VALIDATION OF INTERFERON STIMULATED GENE SCORES, PREVIOUSLY DEVELOPED IN PERIPHERAL BLOOD MONONUCLEAR CELLS, IN WHOLE BLOOD SAMPLES

چکیده

Background A recent EULAR Task force identified critical variability in interferon (IFN) assay methods and defined a research agenda to improve harmonise assays. IFN scores B, developed using stimulated genes (ISG) measured peripheral blood mononuclear cells (PBMC) differentiate between clinical subsets of patients with autoimmune disease [1]. Whole (WB) samples are more commonly used practice trials. Regardless sample type, real time polymerase chain reaction (RT-PCR) experiments require suitable reference gene (RG) which is uniformly expressed across different experimental conditions order normalise target expression quantities [2]. Objectives (i) To determine whether type affects the previously scores; (ii) if so, develop revised WB; (iii) identify best RG for future RT-PCR experiments. Methods Samples were collected from participants enrolled 5 studies; DEFINITION, CONVAS, USEFUL, MASTERPLANs BRAGGSS. PBMCs separated leucosep tubes WB was Tempus™ Blood RNA Tubes. Undetected delta Ct values singly imputed. B calculated paired PBMC agreement types assessed via Bland-Altman (BA) method. New derived exploratory factor analysis (EFA) 31 ISG as per original work Separation subgroups visualized violin plots effect sizes Hedges’ g. identification 16 candidates online software. Results We 342 systemic lupus erythematosus (SLE), 6 ANA positive at risk progressors, 7 non-progressors, 30 rheumatoid arthritis (RA) 10 healthy controls (HC). Subsets comparison (n=45) (n=22). Comparing score versus matched resulted bias -1.44, limits (LOA 0.08, -2.96), spread data on BA plot showed systematic difference. The same gave -1.23 0.26, 2.73), suggested differences not systematic. Mir-183 and FOXO1 gene expression changes in peripheral blood mononuclear cells of breast cancer patients

Background and Aim: Today, cancer is considered as a major health problem and affects the health of society. Breast cancer is the second leading cause of cancer death in women after lung cancer. According to epidemiological studies, cancer is the second most common cause of death after cardiovascular disease worldwide and the third leading cause of death after cardiovascular disease and acciden...

Evaluation of proliferation and cytokines production by mitogen-stimulated bovine peripheral blood mononuclear cells

This in vitro study was conducted to evaluate lymphocyte blastogenic and cytokine production by bovine peripheral blood mononuclear cells (PBMCs) stimulated with phytohemagglutinin (PHA), pokeweed mitogen (PWM) and concanavalin A (Con A) mitogens, by using tetrazolium salt and ELISA tests, respectively. The results presented that  Interleukin-2 (IL-2), IL-4, IL-5, IL-10, IL-17 and IFN-...

Optimization of human peripheral blood mononuclear cells (PBMCs) cryopreservation

Cryopreservation is the method of choice for long term storage of human PBMCs. This study was designed to compare the different combinations of variables affecting the cryopreservation of PBMCs samples. The viability of PBMCs separated from 2×5 ml peripheral blood samples obtained from 16 healthy adult volunteers, were measured using trypan blue dye exclusion method just before freezing with ...

Increased asymmetric dimethylarginine concentrations in stimulated peripheral blood mononuclear cells.

Elevated concentrations of total homocysteine as well as of asymmetric dimethylarginine (ADMA) in the blood have been reported to reflect an increased cardiovascular risk. ADMA is formed by endothelial cells and is an endogenous inhibitor of NO synthase. Earlier we have found that human peripheral blood mononuclear cells (PBMC) produce homocysteine upon stimulation with mitogens concanavalin A,...

Conditioned Medium from Cultured Colorectal Cancer Cells Affects Peripheral Blood Mononuclear Cells Inflammatory Phenotype in Vitro

Background: Colorectal cancer (CRC) is the third most common cancer worldwide. Studies have indicated that immune cells and soluble factors play a key role in maintaining the balance between tumor-promoting inflammation and anti-tumor immunity. It has been shown that secreted cytokines from CRC cell lines could affect peripheral blood mononuclear cells (PBMCs), monocytes, and macrophages phenot...
